What Is Kisspeptin-10?
The KISS1 gene encodes a 145-amino-acid precursor protein cleaved into several active fragments, collectively called kisspeptins, named by length: kisspeptin-54, kisspeptin-14, kisspeptin-13 and kisspeptin-10.
Kisspeptin-10 is the shortest, corresponding to the ten C-terminal residues of the precursor. Critically, it retains full activity at the receptor — the C-terminal region carries the binding and activating determinants, so the shortest fragment is as potent as the longest.
How Researchers Understand Its Mechanism
Kisspeptins act at KISS1R, a G protein-coupled receptor formerly called GPR54. Binding activates Gq and phospholipase C, generating inositol trisphosphate and diacylglycerol and mobilising intracellular calcium — a different route from the cyclic AMP signaling used by many other research peptides.
The functional significance is positional. KISS1R is expressed on GnRH neurons in the hypothalamus, and kisspeptin signaling is now understood as a required upstream regulator of GnRH release. Loss-of-function mutations in KISS1R were found to cause failure of reproductive axis activation, which established the pathway as essential rather than modulatory.
This makes kisspeptin-10 a tool for probing the axis one step above GnRH, and research also examines how kisspeptin neurons integrate signals such as metabolic state and sex steroid feedback.
