PT-141 (Bremelanotide): A Research Overview

PT-141 is a metabolite of Melanotan 2 that differs by a single structural feature, and that difference shifts its receptor selectivity. It is a useful case study in how small modifications alter peptide pharmacology.

What Is PT-141?

PT-141, also known as bremelanotide, is a synthetic cyclic heptapeptide. Structurally it is Melanotan 2 with the C-terminal amide group replaced by a free carboxylic acid — it is in fact a metabolic product of Melanotan 2.

That single change is consequential. Where Melanotan 2 is a broad non-selective agonist across the melanocortin family, PT-141 shows relative preference for MC4R and notably reduced activity at MC1R, the subtype expressed on melanocytes.

How Researchers Understand Its Mechanism

MC4R is expressed predominantly in central nervous tissue, particularly hypothalamic regions, and is one of the most studied melanocortin receptors in neuronal signalling research. PT-141’s relative selectivity for this subtype makes it a more targeted tool than Melanotan 2 for studying MC4R-mediated signaling specifically.

Like other melanocortin receptor agonists, it signals principally through Gs and cyclic AMP. Research has characterised its binding affinity and functional potency across all five subtypes to establish the selectivity profile quantitatively, since “selective” is a relative rather than absolute description.

The Melanotan 2 and PT-141 pair is instructive: because they differ by one terminal modification, comparing them isolates the contribution of that group to receptor discrimination.
